Setting and hooking up a toilet is a surprisingly simple operation.  With an existing fixture, shut off and disconnect the supply line, flush the tank, then swab out remaining water with an old towel or sponge.

If you have a leak behind a tile wall, you'll have to remove all the effected tiles and replace the wall behind the tile.  Adapt the same technique to larger areas, but apply adhesive to the wall, not the tiles.       
Replacing caulk that has deteriorated is important in that you want to prevent any water infiltration along the base of the tub as well as behind the tub walls.  This can lead to costly repairs.
Where do you start?  If you are beginning from scratch with no bath accessories in the bathroom, at most home stores you can find a whole bathroom accessory kit that includes towel racks, tissue holders, and grab bars.
